چکیده

Different formatsthe indirect ELISAs with either peptide or the E7 whole protein as antigens and the more recent GST-E7 capture ELISA have been employed in sero-epidemiological studies for the determination of HPV infection. Further, several studies have attempted to correlate seroprevalence with prognosis and/or early diagnosis of cancer. This study compares the efficacy of ELISA in sero-detection when different forms of the E7 antigen viz., E7 peptides, E7 wholeantigen, and GST-E7 fusion protein are used. The sera samples collected from hospital-visiting women were grouped into two categories“suspect” negative (n=20; from women 18 years of age and negative for HPV 16 L1 antibodies determined by the VLP ELISA) and “suspect” positive (n=40; from women whose corresponding Pap smear samples showed presence of HPV 16 DNA and serum showed sero-positivity for HPV 16 L1 antibodies). The suspect negative sera showed absorbance readings below the respective cut-off points in all formats of the ELISA. However the suspect positive samples showed varied results with ~4%, ~20% and ~73% of samples showing absorbance readings above the cut-off in the E7 Peptide, E7 whole-antigen and GST-E7 capture ELISAs respectively. The results indicate that the GST-E7 based capture ELISA might be best suited for use in the sero detection of antibodies in individuals with unknown disease or infection status.

Transcriptional regulation of E-cadherin and oncoprotein E7 by valproic acid in HPV positive cell lines

Objective(s): Valproic acid (VPA) has proven to be as one of the most promising useful drug with anticancer properties. In this study, we investigate the VPA effects on E-cadherin expression in HeLa, TC1, MKN45, and HCT116 cell lines.  This study assesses the effects of VPA on human papillomavirus E7 expression in HPV positive cell lines. Materials and Methods: Cell lines were treated by2 mmol/...

Developing Michigan Cancer Foundation 7 Cells with Stable Expression of E7 Gene of Human Papillomavirus Type 16

Background: Human papillomavirus (HPV) is responsible for the development of cervical neoplasia.  Infection with human papillomavirus type 16 (HPV-16) is a major risk factor for the development of cervical cancer. The virus encodes three oncoproteins (E5, E6 and E7), of which, the E7 oncoprotein is the major protein involved in cell immortalization and transformation o...

In silico analyzing the molecular interactions of plant-derived inhibitors against E6AP, p53, and c-Myc binding sites of HPV type 16 E6 oncoprotein

Human papillomaviruses (HPV) are a group of strong human carcinogen viruses considered to be the fourth leading cause of mortality among women in the world. HPV is the most important cause of cervical cancer, which is the second most common cancer in women living in low and middle-income countries. To date, there is no effective cure for an ongoing HPV infection; therefore, it is required to in...

Effect of pleurotus sajor-caju polysaccharide encapsulated in poly D, L lactide-co-glycolide nanoparticles for HPV vaccine in murine model

Objective(s): In the current work, poly D, L lactide-co-glycolide (PLGA) particles were applied for a viral vaccine for the delivery of antigens in cytosolic pathway by increasing the antigen presentation to T-lymphocytes. HPV-E7 protein with PLGA particles has been reported as a potent adjuvant for HPV vaccine by encapsulating protein into the PLGA particles. Polysaccharide from Pleurotus sajo...

Detection of human papillomavirus type 18 E7 oncoprotein in cervical smears: a feasibility study.

Persistent infections by high-risk human papillomaviruses (HPVs) are the main etiological factor for cervical cancer, and expression of HPV E7 oncoproteins was suggested to be a potential marker for tumor progression. The objective of this study was to generate new reagents for the detection of the HPV18 E7 oncoprotein in cervical smears. Rabbit monoclonal antibodies against recombinant E7 prot...
